RESUMO

Stressed soft materials commonly present viscoelastic signatures in the form of power-law or exponential decay. Although exponential responses are the most common, power-law time dependencies arise peculiarly in complex soft materials such as living cells. Understanding the microscale mechanisms that drive rheologic behaviors at the macroscale shall be transformative in fields such as material design and bioengineering. Using an elastic network model of macromolecules immersed in a viscous fluid, we numerically reproduce those characteristic viscoelastic relaxations and show how the microscopic interactions determine the rheologic response. The macromolecules, represented by particles in the network, interact with neighbors through a spring constant k and with fluid through a non-linear drag regime. The dissipative force is given by γvα, where v is the particle's velocity, and γ and α are mesoscopic parameters. Physically, the sublinear regime of the drag forces is related to micro-deformations of the macromolecules, while α ≥ 1 represents rigid cases. We obtain exponential or power-law relaxations or a transitional behavior between them by changing k, γ, and α. We find that exponential decays are indeed the most common behavior. However, power laws may arise when forces between the macromolecules and the fluid are sublinear. Our findings show that in materials not too soft not too elastic, the rheological responses are entirely controlled by α in the sublinear regime. More specifically, power-law responses arise for 0.3 ⪅ α ⪅ 0.45, while exponential responses for small and large values of α, namely, 0.0 ⪅ α ⪅ 0.2 and 0.55 ⪅ α ⪅ 1.0.

This work presents a pressure-dependent behavior of silver trimolybdate dihydrate (Ag2Mo3O10·2H2O) nanorods using in situ Raman scattering. The Ag2Mo3O10·2H2O nanorods were obtained by the hydrothermal method at 140 °C for 6 h. The structural and morphological characterization of the sample was performed by powder X-ray diffraction (XRD) and scanning electron microscopy (SEM). Pressure-dependent Raman scattering studies were performed on Ag2Mo3O10·2H2O nanorods up to 5.0 GPa using a membrane diamond-anvil cell (MDAC). The vibrational spectra under high pressure showed splitting and emergence of new bands above 0.5 GPa and 2.9 GPa. Reversible phase transformations under pressure were observed in silver trimolybdate dihydrate nanorods: Phase I - ambient phase (1 atm - 0.5 GPa) â†’ Phase II (0.8 GPa - 2.9 GPa) â†’ Phase III (above 3.4 GPa).

In this work, we present a temperature-dependent behavior of monoclinic silver dimolybdate (m-Ag2Mo2O7) microrods using in situ Raman scattering. The m-Ag2Mo2O7 microrods were obtained by the conventional hydrothermal method at 423 K for 24 h. The structural and morphological characterization of the sample has been done by powder X-ray diffraction (XRD) and scanning electron microscopy (SEM), respectively. Temperature-dependent Raman scattering measurements were performed on m-Ag2Mo2O7 microrods, and the results show an irreversible first-order structural phase transition at 698 K-723 K and the melting process at 773 K. Changes in the Raman spectra confirm the phase transition from the P21/c monoclinic structure to the P-1 triclinic structure. No morphological changes were observed during the structural phase transition of the sample at 723 K. Time-dependent optical microscopy at 773 K showed the growth of nanowires on the Ag2Mo2O7 microrods in the triclinic structure.

Cancer cell lines are widely used as in vitro models of tumorigenesis, facilitating fundamental discoveries in cancer biology and translational medicine. Currently, there are few options for glioblastoma (GBM) treatment and limited in vitro models with accurate genomic and transcriptomic characterization. Here, a detailed characterization of a new GBM cell line, namely AHOL1, was conducted in order to fully characterize its molecular composition based on its karyotype, copy number alteration (CNA), and transcriptome profiling, followed by the validation of key elements associated with GBM tumorigenesis. Large numbers of CNAs and differentially expressed genes (DEGs) were identified. CNAs were distributed throughout the genome, including gains at Xq11.1-q28, Xp22.33-p11.1, Xq21.1-q21.33, 4p15.1-p14, 8q23.2-q23.3 and losses at Yq11.21-q12, Yp11.31-p11.2, and 15q11.1-q11.2 positions. Nine druggable genes were identified, including HCRTR2, ETV1, PTPRD, PRKX, STS, RPS6KA6, ZFY, USP9Y, and KDM5D. By integrating DEGs and CNAs, we identified 57 overlapping genes enriched in fourteen pathways. Altered expression of several cancer-related candidates found in the DEGs-CNA dataset was confirmed by RT-qPCR. Taken together, this first comprehensive genomic and transcriptomic landscape of AHOL1 provides unique resources for further studies and identifies several druggable targets that may be useful for therapeutics and biologic and molecular investigation of GBM.

In this work, we present the study of the temperature-dependent behavior of silver orthophosphate (Ag3PO4) microcrystals using in situ Raman scattering. The Ag3PO4 as-synthesized microcrystals were prepared by the precipitation method and characterized by powder X-ray diffraction (XRD), scanning electron microscopy (SEM), Raman and infrared spectroscopy, and differential scanning calorimetry (DSC). Temperature-dependent phonon dynamics were performed on Ag3PO4 microcrystals and pointed to a first-order phase transition in the temperature range 500-515 °C: Phase I (25-500 °C) â†’ Phase II (515-590 °C). The phase transition is reversible and a temperature hysteresis was observed during the heating - cooling process: Phase II (590-470 °C) â†’ Phase I (455-25 °C). The reversible phase transition is related to the distortion of the tetrahedral symmetry of PO4 caused by the decrease in the crystalline order. DSC analysis confirmed the results of temperature-dependent Raman spectroscopy.

Lidocaine is a versatile drug that not only provides local anesthesia, but also reduces anesthetic requirements of other agents and has antiarrhythmic, pro-kinetic, anti-inflammatory, antiendotoxemic and antioxidant effects. As it is a drug commonly used in critically ill patients, its safety from the cardiovascular system should be ensured. The aim of this study was to determine the effects of a continuous rate infusion (CRI) of lidocaine on left ventricular systolic and diastolic function of healthy rabbits sedated with midazolam by use of transthoracic echocardiography. Ten New Zealand healthy rabbits were sedated with intramuscular midazolam (1 mg/kg) and enrolled in two experimental treatments (control or lidocaine). The control treatment (CT) comprised an intravenous bolus of 0.9% sodium chloride (0.05 mL/kg) followed by CRI at 5 mL/h, whereas the lidocaine treatment (LT) comprised a bolus of 2% lidocaine without epinephrine at 1 mg/kg followed by CRI at 50 µg/kg/minute. Echocardiographic and hemodynamic variables were studied. Variables were recorded at baseline (TB) and 20, 40 and 60 minutes following start of CRI (T20, T40 and T60, respectively). No differences were found between treatments. The results of this study demonstrate that a continuous rate infusion of lidocaine at 50 µg/kg/minute does not impair echocardiographic indices of left ventricular systolic and diastolic function of healthy rabbits sedated with midazolam.

The complex pathophysiology of sickle cell anemia (SCA) involves intravascular hemolytic processes and recurrent vaso-occlusion, driven by chronic vascular inflammation, which result in the disease's severe clinical complications, including recurrent painful vaso-occlusive episodes. Hydroxyurea, the only drug frequently used for SCA therapy, is a cytostatic agent, although it appears to exert nitric oxide/soluble guanylyl cyclase (sGC) modulating activity. As new drugs that can complement or replace the use of hydroxyurea are sought to further reduce vaso-occlusive episode frequency in SCA, we investigated the effects of the sGC agonists BAY 60-2770 (sGC activator) and BAY 41-2272 (sGC stimulator) in the presence or absence of hydroxyurea on SCA vaso-occlusive mechanisms and cell recruitment both ex vivo and in vivo. These agents significantly reduced stimulated human SCA neutrophil adhesive properties ex vivo in association with the inhibition of surface ß2-integrin activation. A single administration of BAY 60-2770 or BAY 41-2272 decreased tumor necrosis factor cytokine-induced leukocyte recruitment in a mouse model of SCA vaso-occlusion. Importantly, the in vivo actions of both agonists were significantly potentiated by the coadministration of hydroxyurea. Erythroid cell fetal hemoglobin (HbF) elevation is also a major goal for SCA therapy. BAY 41-2272 but not BAY 60-2770 at the concentrations employed significantly induced γ-globin gene transcription in association with HbF production in cultured erythroleukemic cells. In conclusion, sGC agonist drugs could represent a promising approach as therapy for SCA, for use either as stand-alone treatments or in combination with hydroxyurea. SIGNIFICANCE STATEMENT: This preclinical study demonstrates that stimulators and activators of sGC are potent inhibitors of the adhesion and recruitment of leukocytes from humans and in mice with sickle cell anemia (SCA) and may represent a promising approach for diminishing vaso-occlusive episode frequency in SCA. Hydroxyurea, a drug already frequently used for treating SCA, was found to potentiate the beneficial effects of sGC agonists in in vivo studies, implying that these classes of compounds could be used alone or in combination therapy.

We study, numerically, a system of active particles with either a single noise value or a mixture of equal proportions of particles with two noise values under the influence of an attractive periodic background potential, and we observe their diffusion regimes and trapping states. For the single noise system, we show that the slow diffusion is correlated to a significant particle trapping, while normal diffusion is seen for partial or no trapping. Our results indicate that low noise particles are less susceptible to the background, i.e., they have a smaller chance to be trapped as compared to higher noise particles for the same background, and that denser systems achieve a no-trapping state, unless for the largest noise value we studied. For the mixtures, we study the sorting of particles based on their noise value differences and observe that particles with distinct noises are trapped at distinct radii compared to a trap minimum, and, since these radii depend on the density, the latter should be well tuned in order to have an efficient sorting.

We investigate the transport of active matter in the presence of a disordered square lattice of asymmetric obstacles, which is built by removing a fraction of them from the initial full lattice. We obtain a spontaneous inversion of the net particle current, compared to the usual sense of such a current as a function of the fraction of removed obstacles and particle density. We observed that the negative current regime is the consequence of trapping of particles among the obstacles which favors that more particles move in the negative current direction. The same reasoning applies to the positive current regime as well. We show a calculation that partially reproduces our numerical results, based on the argument that the mean current is given by the product of the mean speed and the mean number of travelers in each direction; the breakdown of this assumption is responsible for the failure of our calculation to reproduce the initial negative current regime.

We investigate a two-dimensional system of magnetic colloids with anisotropic geometry (rods) subjected to an oscillating external magnetic field. The structural and dynamical properties of the steady states are analyzed, by means of Langevin dynamics simulations, as a function of the misalignment of the intrinsic magnetic dipole moment of the rods with respect to their axial direction, and also in terms of the strength and rotation frequency of an external magnetic field. The misalignment of the dipole relative to their axial direction is inspired by recent studies, and this is extremely relevant in the microscopic aggregation states of the system. The dynamical response of the magnetic rods to the external magnetic field is strongly affected by such a misalignment. Concerning the synchronization between the magnetic rods and the direction of the external magnetic field, we define three distinct regimes of synchronization. A set of steady states diagrams are presented, showing the magnitude and rotation frequency intervals in which the distinct self-organized structures are observed.

PURPOSE: Coronary artery geometry can have a significant impact in the hemodynamic behavior of coronary blood flow, influencing atherosclerotic plaque formation. The present work focuses on, through a statistical study, the connection between several geometric parameters of the right coronary artery-ostium cross-sectional area, angles between the common trunk and the side-branches, tortuosity, curvature and cross-sectional area in each side-branch-and their influence on hemodynamic descriptors. Parameters such as low wall shear stress and local disturbed flow, which are associated with atherosclerosis formation, were analysed. METHODS: Computed tomography images of ten healthy individuals were selected to reconstruct in vivo three-dimensional models of right coronary arteries. Blood flow was simulated through a compliant model with realistic boundary conditions. Calculated hemodynamic descriptors values were correlated with the geometric parameters using the Pearson correlation coefficient (r) and the p value. RESULTS: The strongest correlations were found in the middle and distal segments of the right coronary artery. A decrease in the ostium area promotes a decrease in the WSS magnitude from the proximal to the distal segment (r = 0.82). Very strong correlations (r > 0.90) were achieved between geometric parameters (cross-sectional area, angle, tortuosity) of the right-ventricular branch and the wall shear stress magnitude in the middle and distal segments. CONCLUSIONS: Low values of tortuosity, smaller cross-sectional area and higher angle of the right-ventricular branch leads to a hemodynamic behavior more propitious to atherosclerosis formation, within the study cases. The right-ventricular branch seems to have the highest influence in the hemodynamic behavior of the right coronary artery.

Bacillus species are becoming increasingly relevant for use as probiotics or feed additives where their heat stability can ensure survival in the food matrix or enable long-term storage at ambient temperature. Some Bacillus species are pigmented and in this study, we have examined two strains, one Bacillus pumilus (pigmented red) and the other Bacillus megaterium (pigmented yellow) for their safety for potential use in humans as dietary supplements. In addition, we have set out to determine if they might confer any potential health benefits. Both strains produce C30 carotenoids while the B. pumilus strain also produced large quantities of riboflavin equivalent to genetically modified Bacillus strains and most probably contributing to this strain's pigmentation. Riboflavin's and carotenoids are antioxidants, and we have evaluated the ability of vegetative cells and/or spores to influence populations of Faecalibacterium prausnitzii in the colon of mice. While both strains increased levels of F. prausnitzii, spores of the B. pumilus strain produced a significant increase in F. prausnitzii levels. If found to be reproducible in humans such an effect might, potentially, confer health benefits particularly for those suffering from inflammatory bowel disease.

Objectives: Gonorrhoea and antimicrobial resistance (AMR) in Neisseria gonorrhoeae are major public health concerns globally. Enhanced AMR surveillance for gonococci is essential worldwide; however, recent quality-assured gonococcal AMR surveillance in Latin America, including Brazil, has been limited. Our aims were to (i) establish the first nationwide gonococcal AMR surveillance, quality assured according to WHO standards, in Brazil, and (ii) describe the antimicrobial susceptibility of clinical gonococcal isolates collected from 2015 to 2016 in all five main regions (seven sentinel sites) of Brazil. Methods: Gonococcal isolates from 550 men with urethral discharge were examined for susceptibility to ceftriaxone, cefixime, azithromycin, ciprofloxacin, benzylpenicillin and tetracycline using the agar dilution method, according to CLSI recommendations and quality assured according to WHO standards. Results: The levels of resistance (intermediate susceptibility) to tetracycline, ciprofloxacin, benzylpenicillin and azithromycin were 61.6% (34.2%), 55.6% (0.5%), 37.1% (60.4%) and 6.9% (8.9%), respectively. All isolates were susceptible to ceftriaxone and cefixime using the US CLSI breakpoints. However, according to the European EUCAST cefixime breakpoints, 0.2% (n = 1) of isolates were cefixime resistant and 6.9% (n = 38) of isolates had a cefixime MIC bordering on resistance. Conclusions: This study describes the first national surveillance of gonococcal AMR in Brazil, which was quality assured according to WHO standards. The high resistance to ciprofloxacin (which promptly informed a revision of the Brazilian sexually transmitted infection treatment guideline), emerging resistance to azithromycin and decreasing susceptibility to extended-spectrum cephalosporins necessitate continuous surveillance of gonococcal AMR and ideally treatment failures, and increased awareness when prescribing treatment in Brazil.

Based on extensive Langevin Dynamics simulations we investigate the structural properties of a two-dimensional ensemble of magnetic rods with a peapod-like morphology, i.e, rods consisting of aligned single dipolar beads. Self-assembled configurations are studied for different directions of the dipole with respect to the rod axis. We found that with increasing misalignment of the dipole from the rod axis, the smaller the packing fraction at which the percolation transition is found. For the same density, the system exhibits different aggregation states for different misalignment. We also study the stability of the percolated structures with respect to temperature, which is found to be affected by the microstructure of the assembly of rods.

Two-dimensional systems of inverse patchy colloids modeled as disks with a central charge and having their surface decorated with oppositely pointlike charged patches are investigated using molecular dynamics simulations. The self-assembly of the patchy colloids leads to diverse ground state configurations ranging from crystalline arrangements of monomers to linear clusters, ramified linear clusters and to percolated configurations. Two structural phase diagrams are constructed: (1) as a function of the net charge and area fraction, and (2) as a function of the net charge and the range of the pair interaction potential. An interesting reentrant percolation transition is obtained as a function of the net charge of the colloids. We identify distinct mechanisms that lead to the percolation transition.

The objective of this study was to evaluate the influence of collard green stalks as environmental enrichment on the behavior of cockatiels (Nymphicus hollandicus) maintained in captivity. Sixteen birds were housed individually in cages and fed a commercial diet for parrots and seed mix, with water ad libitum during 12 days in climatic chamber with temperature and relative humidity simulating tropical conditions. The animals (n = 8) were divided into two groups, control and enriched, and the experimental period was divided into two phases (during and after enrichment) of four days each. The collard green stalks were used as enrichment only during the first period of the experiment, daily changed and maintained only during the day. The stalks increased the feed intake and decreased the sleep activities. No effect was observed on the body surface temperature, locomotion, maintenance and other resting activities. Small undesirable activities were detected. It was concluded that collard green stalks can be used as environmental enrichment, becoming part of food preferences of cockatiels. However, its use does not significantly alter the behavior of birds kept in captivity.(AU)


O objetivo deste estudo foi avaliar a influência de talos de couve como enriquecimento ambiental sobre o comportamento de calopsitas (Nymphicus hollandicus) mantidas em cativeiro. Dezesseis aves foram alojadas individualmente em gaiolas e alimentadas com dieta comercial para psitacídeos e mistura de sementes, com água ad libitum durante 12 dias, em câmara climática com temperatura e umidade relativa do ar simulando condições tropicais. Os animais (n=8) foram divididos em dois grupos, controle e enriquecido, e o período experimental foi dividido em duas fases (durante e após o enriquecimento) de quatro dias cada. Os talos de couve foram usados como enriquecimento apenas durante o primeiro período do experimento, trocados diariamente e mantidos apenas durante o dia. Os talos aumentaram o consumo de ração e diminuíram o comportamento dormir. Nenhum efeito foi observado nas outras atividades de descanso, na temperatura da superfície corporal, na locomoção e manutenção. Foram detectadas pequenas atividades indesejáveis. Concluiu-se que os talos de couve podem ser usados como enriquecimento ambiental, formando parte das preferências alimentares de calopsitas. No entanto, seu uso não altera de maneira expressiva o comportamento das aves.(AU)

We investigated the hypothesis that the increased concentration of plasma methylguanidine (MG) increases oxidative metabolism and accelerates apoptosis of neutrophils from dogs with chronic kidney disease (CKD). To achieve this, the levels of MG were quantified in healthy (n=16) and uremic dogs with CKD stage 4 of according to the guidelines of the International Renal Interest Society (IRIS, 2015) (n=16) using high performance liquid chromatography (HPLC). To evaluate the isolated effect of MG on neutrophil oxidative metabolism and apoptosis, neutrophils isolated from 12 healthy dogs were incubated with the highest concentration of plasma MG (0.005g/L) observed in dogs with CKD. Neutrophil oxidative metabolism was assessed by flow cytometry, using the probes hydroethidine for superoxide production and 2',7'-dichlorofluorescein diacetate for hydrogen peroxide production, with or without phorbol myristate acetate (PMA) stimulus. Neutrophil apoptosis and viability were also evaluated in flow cytometer using the Annexin V-PE system, with or without the apoptosis-inducing effect of camptothecin. Uremic dogs presented higher concentrations of MG (p<0.0001), increased oxidative stress and primed neutrophils with higher apoptosis rate. The neutrophil abnormalities observed in vivo were also reproduced in vitro, using cells isolated from healthy dogs and incubated with MG. We obtained strong evidence that in dogs with CKD, increased MG levels contributed to oxidative stress and potentially compromised the non-specific immune response by altering the oxidative metabolism and viability of canine neutrophils.

We present a theoretical study of the one-electron states of a semiconductor-made quantum ring (QR) containing a series of piecewise-constant wells and barriers distributed along the ring circumference. The single quantum well and the superlattice cases are considered in detail. We also investigate how such confining potentials affect the Aharonov-Bohm like oscillations of the energy spectrum and current in the presence of a magnetic field. The model is simple enough so as to allow obtaining various analytical or quasi-analytical results. We show that the well-in-a-ring structure presents enhanced localization features, as well as specific geometrical resonances in its above-barrier spectrum. We stress that the superlattice-in-a-ring structure allows giving a physical meaning to the often used but usually artificial Born-von-Karman periodic conditions, and discuss in detail the formation of energy minibands and minigaps for the circumferential motion, as well as several properties of the superlattice eigenstates in the presence of the magnetic field. We obtain that the Aharonov-Bohm oscillations of below-barrier miniband states are reinforced, owing to the important tunnel coupling between neighbour wells of the superlattice, which permits the electron to move in the ring. Additionally, we analysis a superlattice-like structure made of a regular distribution of ionized impurities placed around the QR, a system that may implement the superlattice in a ring idea. Finally, we consider several random disorder models, in order to study roughness disorder and to tackle the robustness of some results against deviations from the ideally nanostructured ring system.
